1,2-Dioleoyl-sn-glycero-3-PG (sodium salt) is an anionic phospholipid that plays a significant role in the structure and function of cell membranes[1]. 1,2-Dioleoyl-sn-glycero-3-PG (sodium salt) is wildly used in the preparation of liposomes and other artificial membrane systems for studying the behavior of membrane proteins[2], drug delivery mechanisms[3][4], and the structure and function of cell membranes[5].

1,2-Dioleoyl-sn-glycero-3-PG (sodium salt) can suppress the production of inflammatory mediators induced by recombinant S100A9 through both TLR2 and TLR4 pathways. In RAW264.7 cells, treatment with 100mg/ml DOPG and 2.5mg/ml S100A9 for 2 hours significantly inhibit the expression of IL-1β, IL-6, and TNF-α (tumor necrosis factor-α)[6].

1,2-Dioleoyl-sn-glycero-3-PG (sodium salt) in water (prepared by sonicating a 1,2-Dioleoyl-sn-glycero-3-PG (sodium salt) lipid film in deionized water) applied to the imiquimod (IMQ) treated mouse skin can inhibited IMQ-induced psoriasiform phenotype such as ear thickness and epidermal hyperplasia[6].

Preparation Method

RAW264.7 cells were treated with 100mg/ml 1,2-Dioleoyl-sn-glycero-3-PG(sodium salt) and 2.5mg/ml S100A9 for 2 hours. The expression level of the inflammatory mediators IL-1b, IL-6, and TNF-a were measured by quantitative real-time reverse transcriptase-PCR with GAPDH used as the housekeeping gene.

Preparation Method

Mice were treated with vehicle (petroleum jelly) or IMQ daily for 5 days in the morning. In the afternoon, water or 1,2-Dioleoyl-sn-glycero-3-PG(sodium salt) solution was applied to the treated skin. Then the Psoriasis Area and Severity Index score was estimated in a blinded manner.
