Description

BODIPY-aminoacetaldehyde diethyl acetal (BAAA-DA) is a stable precursor of BODIPY-aminoacetaldehyde (BAAA), a cell-permeable fluorescent substrate for aldehyde dehydrogenase (ALDH) with an Ex/Em of 488/512nm[1-4].

BODIPY-aminoacetaldehyde diethyl acetal converts to BAAA in acidic conditions. BAAA is uncharged and can diffuse freely across the plasma membrane of intact living cells with cellular permeability. BAAA is converted into Bodipy-amino acetic acid (BAA) intracellular by ALDH, which is retained by cells due to the net negative charge of BAA and can be used to identify cells with high ALDH activity [1-2]. BODIPY-aminoacetaldehyde diethyl acetal has been used to isolate human hematopoietic progenitor cells, which have high ALDH activity, and via flow cytometry to sort cancer stem cells that contain high levels of ALDH[1,3]. BODIPY-aminoacetaldehyde diethyl acetal can be used to characterize live ALDHpos cells, and the BAA used in the cells can be excited at 488nm with a maximum emission wavelength of 512nm[4].

BODIPY-aminoacetaldehyde diethyl acetal在酸性条件下转化为BAAA。BAAA不带电荷,可以在完整活细胞的质膜上自由扩散,具有细胞渗透性。BAAA通过ALDH在细胞内转化为BODIPY-氨基乙酸(BAA),由于BAA净负电荷,BAA被细胞保留并可用于识别具有高ALDH活性的细胞[1-2]。BODIPY-aminoacetaldehyde diethyl acetal已用于分离具有高ALDH活性的人造血祖细胞并通过流式细胞术分选含有高水平ALDH的癌症干细胞[1,3]。BODIPY-aminoacetaldehyde diethyl acetal可用于鉴定活ALDHpos细胞,可以在488nm处激发,最大发射波长为512nm[4]

实验参考方法

本实验方案来自文献,请根据具体需要进行修改。

1、制备BODIPY-aminoacetaldehyde diethyl acetal染色液

(1)配制BODIPY-aminoacetaldehyde diethyl acetal染料储存液:使用DMSO溶解固体粉末,配置成BODIPY-aminoacetaldehyde diethyl acetal染料储存液。

注意:BODIPY-aminoacetaldehyde diethyl acetal储存液建议分装后于-4℃或-20℃避光保存,避免反复冻融。

(2)工作液制备:加入等体积的2N HCl将BODIPY-aminoacetaldehyde diethyl acetal转化为BAAA,2小时后,加入合适的缓冲液(如PBS),配制浓度为1-20μM的BODIPY-aminoacetaldehyde diethyl acetal工作液。

注意:请根据实际情况调整BODIPY-aminoacetaldehyde diethyl acetal工作液浓度,现用现配。

2、细胞染色

悬浮细胞:悬浮细胞经1000g离心3-5分钟。弃去上清液,使用PBS清洗两次,每次5分钟。

贴壁细胞:加入胰蛋白酶消化细胞,1000g离心3-5分钟。弃去上清液,使用PBS清洗两次,每次5分钟。

(1)在收取的细胞中加入1mL的BODIPY-aminoacetaldehyde diethyl acetal工作液,37℃避光孵育30-60分钟。

(2)孵育结束后,经1000g离心5分钟,去除上清液,加入PBS清洗2-3次,每次5分钟。

(3)使用PBS重悬细胞,通过流式细胞仪进行分析(Ex/Em为488/512nm)。

注意事项:

①本品为未活化的荧光染料,需先使用酸(2N HCl)进行活化;

②荧光染料均存在淬灭问题,请尽量注意避光;
