Endothelin 1 (swine, human) (TFA) is a synthetic peptide with human and porcine Endothelin 1 (ET-1) sequences and is a potent endogenous vasoconstrictor[1]. Endothelin 1 acts through ETA and ETB receptors and has proinflammatory and profibrotic effects[2]. Endothelin 1 is a potent vasoconstrictor and mitogen that contributes to elevated blood pressure and associated vascular remodeling and target organ damage[3]. Endothelin 1 can act as an autocrine/paracrine growth factor in porcine ovarian granulosa cells and human cancer cell lines[4, 5].
In vitro, treatment of porcine ovarian (granulosa) cell monolayers with Endothelin 1 (swine, human) (TFA) (100nM) rapidly increased intracellular soluble inositol polyphosphate content, elevated Ca2+ concentration, and inhibited follicle-stimulating hormone (FSH)-stimulated progesterone and cAMP accumulation[6].

Preparation Method | Swine ovarian (granulosa) cells monolayer cultures were established in M199 (which contains 0.05mg/liter inositol) supplemented with 3% FBS and porcine insulin (3pg/mL) at an initial plating density of 1×107 viable tells/35-mm tissue culture. After 48h, the incubation medium was exchanged with fresh serum-free M199 supplemented with 0.1% BSA. After 24h, cells were prelabeled with [3H]myoinositol dishes by exposing them to M199 containing 0.1% BSA and 2.5μCi/mL [3H]myoinositol for 18h. Thereafter, cells were washed twice in M199 with 0.1% BSA and incubated for 1h in the presence of 10mM unlabeled inositol. During the last 15min of this “cold chase” period, 30μL of a 0.5M stock LiCl solution were added to each dish ( final concentration, 10mM). At designated intervals, medium alone (control) or Endothelin 1 (final concentration, 100nM) was added in 80μL aliquots. The reaction was stopped by the addition of ice-cold chloro- form-methanol (1:2, vol/vol). Samples were maintained on ice for about 1h while cells were scraped from the culture dishes and centrifuged in bio-vials. [3H]Inositol phosphates were separated by anion exchange chromatography. |
