Description

Fast Red Violet LB is an azo dye that can be used to detect tartrate-resistant acid phosphatase (TRAP) and alkaline phosphatase (ALP)[1, 2]. Fast Red Violet LB, also known as carminic acid, is extracted from the scales of female cochineal insects[3]. Fast Red Violet LB can be used to study the role of TNF-α in fracture healing[4]. Fast Red Violet LB can stain naphthol AS-D chloroacetate esterase activity and can be used to detect granulocytes[5].

实验参考方法

本方案仅提供一个指导,请根据您的具体需要进行修改。

1. 溶液配制

(1)工作液:用实验缓冲液配制Fast Red Violet LB染色液,配置浓度为0.6mg/mL。

注意:最佳的工作浓度请根据实际情况调整或参阅文献自行设置梯度浓度进行摸索。工作液必须现配现用。

2. 用Fast Red Violet LB进行抗酒石酸酸性磷酸酶(TRAP)染色(来自文献,仅做参考)

(1)细胞染色:将细胞用PBS中的4%多聚甲醛固定,然后用PBS洗涤,并用丙酮和乙醇(1:1)的混合物处理1min,再次用PBS洗涤,并在37°C下在含有0.2mg/mL萘酚AS-MX磷酸盐和0.6mg/mL Fast Red Violet LB的TRAP缓冲液(pH 5.2)中孵育30min。用纯水洗涤后,在显微镜下观察[1]

(2)组织染色:将载玻片(股骨样品)在含有萘酚AS-MX磷酸盐、Fast Red Violet LB和MnCl2的乙酸钠缓冲液(0.1M,pH 5.0)中在酒石酸钠存在下于37 ℃孵育60min,用于TRAP染色[2]

3. 用Fast Red Violet LB进行碱性磷酸酶(ALP)染色(来自文献,仅做参考)

(1) 细胞染色:细胞固定后用0.1mg/mL萘酚AS-MX磷酸盐和0.6mg/mL Fast Red Violet LB对细胞进行染色,然后用数码相机拍照[3]

(2) 细胞3D培养物染色:用冷的10%中性福尔马林缓冲液固定样品15min。用dH2O冲洗后,在室温下,将样品与0.1mg/mL萘酚AS-MX磷酸盐底物(溶于0.4% N,N-二甲基甲酰胺和0.1M HCl,pH 8.3,含0.6mg/mL Fast Red Violet LB)孵育45min,以使产物可视化。染色液用dH2O洗去,在立体镜下观察,使用ImageJ软件通过测量每张图像中红色染色面积与总面积的百分比来定量ALP染色[4]
