PC DBCO-PEG4-NHS ester is a PEG based PROTAC linker that can be used as a linker in click chemistry for the synthesis of PROTAC molecules. The reaction between the DBCO group in PC DBCO-PEG4-NHS ester and azide compounds (usually azide methane (CH2N2) or azide amide analogues) belongs to the stress promoted azide alkyne cycloaddition reaction (SPAAC), which is a metal free chemical reaction. The alkyne portion of DBCO can bind with azide groups and form a stable 1,2,3-triazole cyclobutane compound through a [3+2] cycloaddition mechanism.
PC DBCO-PEG4-NHS ester can be used for surface modification of samples such as peptides, proteins, enzymes, and live cells, to prepare protein peptide conjugates, protein/peptide oligonucleotide conjugates, and antibody enzyme/antibody drug conjugates. PC DBCO-PEG4-NHS ester only reacts with azide compounds, exhibiting good bioorthogonality, high selectivity, and high reaction rate. Due to the absence of metal catalysts in the reaction, this product is also suitable for use in living organisms or other metal sensitive environments, and has broad application prospects and scientific value.
It is recommended to explore the labeling ratio of PC DBCO-PEG4-NHS ester to ligand at around 5:1 or 8:1, and label at room temperature for 30-1h. The usage conditions need to be adjusted according to specific experimental needs and labeling effects.
