Sesquicillin A is a fungal metabolite that has been found in Albophoma.1 It inhibits the growth of A. salina brine shrimp (MIC = 6.25 µg/ml). Sesquicillin is cytotoxic to Jurkat cells (IC50 = 34 µM). It induces cell cycle arrest at the G1 phase and inhibits proliferation of MCF-7 breast cancer cells when used at a concentration of 20 µg/ml.2

Divergent Synthesis of Pyrone Diterpenes via Radical Cross Coupling

A divergent strategy for assembling pyrone diterpenes is presented. Capitalizing on the unique stereo- and chemoselectivity features of radical-based chemistry, the core decalin of these structures is efficiently forged using an electrochemically assisted oxidative radical polycyclization while key peripheral substituents are appended using decarboxylative radical cross couplings. In this way, access to four natural products (subglutinols A/B, higginsianin A, and Sesquicillin A) is achieved in a concise and stereocontrolled fashion that is modular and amenable to future medicinal chemistry explorations.

Four new antibiotics, sesquicillins B to E were isolated from the culture broth of Albophoma sp. FKI-1778 together with known sesquicillin (Sesquicillin A in this paper). The structures of sesquicillins were elucidated by spectroscopic studies including various NMR experiments. All sesquicillins have a common pyrano-diterpene skeleton. Sesquicillins showed moderate inhibitory activity against the growth of Artemia salina (brine shrimps) and Jurkat cells.
